Full-Length Isoform Sequencing (Iso-seq) refers to the application of PacBio SMRT sequencing technology to obtain completed sequences of RNA molecules (from the 5’ end of transcripts to the 3’ end poly-A tail). The Iso-seq method does not require fragmentation of RNA molecules, thus enabling the generation of full-length sequences without assembly. The Iso-seq method provides more accurate information of transcripts, helping to identify more alternative splicing events, novel genes and transcript isoforms, as well as fusion genes.
